Association of LRP5 gene polymorphisms rs4988331 and rs599083 with osteoporosis in postmenopausal women

Abstract
Objective To evaluate the association between polymorphisms at rs4988331 and rs599083 loci of the low-density lipoprotein receptor-related protein 5(LRP5)gene and postmenopausal osteoporosis(OP)in women in Shihezi,Xinjiang and to provide a scientific basis for the prevention and treatment of OP.Methods Participants were classified into three groups based on bone mineral density(BMD)measured by dual energy X-ray absorptionmetry(DXA):normal bone mass group(n=57),osteopenia group(n=53),and OP group(n=32).Data collected included age,years since menopause,and body mass index(BMI).Clinical biochemical indicators of glucose,lipid,and bone metabolism were measured.Single nucleotide polymorphisms(SNPs)at LRP5 gene loci were detected using mass spectrometry.Results(1)Compared to the normal bone mass group,the osteopenia and OP groups had signifi-cantly higher age and years since menopause(P<0.01).(2)Compared to the normal bone mass group,the osteope-nia group had higher glycosylated hemoglobin(HbA1c)(P<0.05).The OP group showed significantly lower triglycer-ide(TG),triglyceride-glucose(TyG)index,TyG-BMI index(P<0.05).Compared to the osteopenia group,the OP group had lower TG,TyG-BMI index(P<0.05).(3)There were no significant differences in type 2 diabetes mellitus(T2DM)prevalence or polymorphism distributions of rs4988331 and rs599083 among the groups(P>0.05).(4)In the normal bone mass group,individuals with the CC genotype at rs4988331 had higher fasting blood glucose(FBG)and lower serum creatinine(Cr)compared to those with the CT genotype(both P<0.05).In the abnormal bone mass group,the CC genotype was associated with higher FBG,HbA1c,L1-4 BMD,and TyG-waist circumference(WC)in-dex(all P<0.05).In the normal bone mass group,individuals with the TT genotype at rs599083 had higher low-densi-ty lipoprotein cholesterol(LDL-C)but lower BMD at L1-4 and FN than those with GG/GT genotypes(all P<0.05).(5)Increased age was a risk factor for decreased BMD at L1-4 and FN(P<0.05),while a higher TyG-BMI index was a protective factor for BMD loss at FN and L1-4.Conclusion In postmenopausal women in Xinjiang,SNPs at the rs4988331 locus of the LRP5 gene is associated with OP and T2DM,while SNPs at rs599083 is associated with OP de-velopment.关键词
