Identification and Analysis of Microsatellites in Three Strains of Vero Cells for Animal Quarantine

Abstract
To clarify the genetic characteristics and potential cross-contamination of three Vero cell lines preserved in the laboratory,this study employed microsatellite(STR)technology for molecular biological identification.DNA was extracted from the three Vero cell lines,followed by multiplex amplification of 10 loci,including 8 monkey/human-derived loci(such as D8S1106),1 rhesus monkey-specific locus(F13A01),and 1 human-specific locus(FGA).PCR products were detected and analyzed using an ABI 3730xl Genetic Analyzer.The results showed that the STR typing of two Vero cell lines fully matched the standard Vero cell line without contamination,while one cell line exhibited abnormal peaks at D19S245 and D6S1017 loci,indicating potential genetic variation.The STR identification method established herein can effectively verify the identity of Vero cells and detect contamination,providing technical support for constructing cell banks in animal quarantine.关键词
