雷公藤红素对口腔鳞状细胞癌细胞恶性生物学表型的影响及作用机制研究
Effects of tripterine on the malignant biological phenotype of oral squamous cell carcinoma and its mechanism of action

Abstract
Objective To investigate the effect of tripterine on the malignant biological phenotype of oral squamous cell carcinoma(OSCC)cells and analyze the possible mechanism of action.Methods OSCC SCC-25 cells were used as a model.The cells were treated with different concentrations of tripterine(low dose 100 mg/L,medium dose 200 mg/L,and high dose 400 mg/L).The control group was treated with 0.1%dimethyl sulfoxide.After 72 h of treatment,the cell proliferation rate was detected using a cell counting kit-8,and the cell colony formation ability was determined using a colony formation assay.The cell apoptosis rate was determined by flow cytometry,and the cell cycle distribution was analyzed using propidium iodide staining.The cell invasion and migration abilities were evaluated using Transwell and wound healing assays.The mRNA and protein expression level changes of microRNA-24-3p(miR-24-3p),F-box and WD Repeat Domain Containing 7(FBXW7),and sterol regulatory element-binding protein 1(SREBP-1)were detected by qRT-PCR and Western blotting.Results Compared with the control group,after tripterine intervention,the proliferation rate,number of monoclonal formation,invasion ability(number of cells penetrating the membrane),and migration rate of SCC-25 cells in all groups were significantly decreased,while the apoptosis rate and G2/M phase ratio were significantly increased.The expression levels of miR-24-3p and SREBP-1 mRNA and protein were downregulated,while the expression levels of FBXW7 mRNA and protein were upregulated,with statistically significant differences(all P<0.05).All indicators changed in a dose-dependent manner with increasing tripterine concentration,the differences of which were statistically significant(P<0.05).Conclusion Tripterine may inhibit miR-24-3p expression and activate FBXW7-mediated ubiquitination and degradation of SREBP-1,thereby targeting and regulating multiple malignant phenotypes of OSCC cells,including proliferation,apoptosis,invasion,and lipid metabolism reprogramming.关键词
