NF-κB/IL-6/STAT3信号通路对结直肠腺瘤性息肉的作用研究
Research on the effects of the NF-κB/IL-6/STAT3 signaling pathways on adenomatous polyps

Abstract
Objective To explore the differences in expression of the nuclear factor-κB(NF-κB)/interleukin-6(IL-6)/signal transducer and activator of transcription 3(STAT3)signaling pathways in colorectal adenomatous polyps and hyperplastic polyps,and analyze its potential role in the occurrence and development of intestinal tumors.Methods Polyp tissue specimens from 3 patients each with colorectal adenomatous polyps and hyperplastic polyps who underwent endoscopic polyp removal at Hangzhou Third People's Hospital from April to June 2024 were retrospectively selected,as well as mucosal tissue specimens of colorectal inflammation from 3 patients with colorectal mucosal inflammation.The expression levels of NF-κB,IL-6,and STAT3 mRNA and protein were measured using Western blotting and quantitative real-time PCR(qRT-PCR),and the expression levels were compared among the different tissue types.Results No statistically significant differences were found in NF-κB and IL-6 mRNA levels among colorectal adenomatous polyps,hyperplastic polyps and inflammatory mucosal tissues(all P>0.05).However,STAT3 mRNA expression level showed significant differences(P<0.05).Specifically,STAT3 mRNA level was higher in adenomatous polyp tissues compared to inflammatory mucosal tissues(P<0.05).There were statistically significant differences in the protein expression levels of NF-κB,IL-6 and STAT3 among colorectal adenomatous polyps,hyperplastic polyps and inflammatory mucosal tissues(all P<0.05).Pairwise comparisons between groups revealed higher protein levels of STAT3,NF-κB,and IL-6 in adenomatous polyp tissues than those in inflammatory mucosal tissues(all P<0.05).Differences between other groups were not statistically significant(all P>0.05).Conclusion NF-κB,IL-6,and STAT3 might promote the occurrence and development of colorectal adenomatous polyps.Consequently,targeted studies on the NF-κB/IL-6/STAT3 signaling pathway could provide new treatment options for colorectal tumors.关键词
