头针对脑出血后脑组织内前列腺素内过氧化物合成酶2调控的实验研究
A study on the experiment about regulating PTGS2 in brain tissue after cerebral hemorrhage by scalp acupuncture

Abstract
Objective:To investigate the effects of acupuncture at Baihui acupoint(DU20)penetrating Qubin acupoint(GB7)on the recovery of neurological function,nerve cell apoptosis,and expression of PTGS2 in rats with intracerebral hemorrhage(ICH).Methods:A total of 50 μL of autologous blood was injected into the caudate nucleus of rat right brain to make ICH model.After successful modeling,randomly selected 18 successfully modeled rats and divided into model group,acupuncture group,and sham acupuncture group,with 6 rats in each group.Then selected several rats and injected 10 µL(0.1 µg/mL)RSL3 into the right brain.Thirty minutes later,ICH models were established.After awakening,randomly selected 12 successfully modeled rats and divided them into agonist group and agonist+acupuncture group,with 6 rats in each group.Finally,6 healthy rats were randomly selected and divided in blank group.After grouping,rats in blank group,model group,and agonist group were not treated.Rats in acupuncture group and agonist+acupuncture group were treated with daily Baihui-penetrating-Qubin acupuncture.The sham acupuncture group inserted needle horizontally 1 cm back of Baihui-penetrating-Qubin acupuncture.After 7 consecutive days of treatment,the Bederson neurological function score was used to evaluate the neurological function of rats before and after modeling.TUNEL assay was used to detect the level of nerve cell apoptosis in rat brain,and Western blot was used to detect the expression of PTGS2 in rat brain.Results:①Acupuncture at Baihui acupoint penetrating Qubin acupoint can significantly improve the neurological function of ICH rats:Bederson score in the acupuncture group was decreased significantly at the 7-day time point compared with the 1-day time point within group(P<0.001).At the 7-day time point,the Bederson score in the acupuncture group was decreased significantly compared with the model group,with a statistically significant difference(P<0.05).The Bederson score in the acupuncture group at 7-day time point was significantly lower than that in the non-meridian non-acupoint group,with a statistically significant difference(P<0.001).②Acupuncture at Baihui acupoint penetrating Qubin acupoint could significantly reduce the content of apoptotic cells in the brain tissue of ICH rats:the number of TUNEL positive cells in the acupuncture group was less than that in the model group,with a statistically significant difference(P<0.05).The number of TUNEL positive cells in the acupuncture group was less than that in the non-meridian non-acupoint group,with a statistically significant difference(P<0.05).③Acupuncture at Baihui acupoint penetrating Qubin acupoint could significantly downregulate the expression of PTGS2 protein in the brain tissue of ICH rats:the PTGS2 protein expression in the acupuncture group was lower than that in the model group,with a statistically significant difference(P<0.05).The expression of PTGS2 protein in the acupuncture group was lower than that in the non-meridian non-acupoint group,with a statistically significant difference(P<0.05).Conclusions:Acupuncture at Baihui acupoint penetrating Qubin acupoint can significantly promote the recovery of neurological function in ICH rats,which may be related to its inhibition of PTGS2 protein expression in brain tissue,thereby inhibiting neuronal apoptosis.关键词
