针刺对脑出血后脑组织铁重链蛋白1影响的实验研究
An experimental study on the influence of acupuncture on the expression of FTH1 in brain tissue after intracerebral hemorrhage

Abstract
Objective:To investigate the influences of acupuncture at"Baihui"acupoint(GV20)-penetrating-"Qubin"acupoint(GB7)on the recovery of neurological function,mitochondrial destruction,and the expression of ferritin heavy 1(FTH1)in rats with intracerebral hemorrhage(ICH).Methods:Fifty μL of tail autologous blood was injected into the caudate nucleus of rat right brain to establish ICH model.Bederson neurological function score was used to select successful ICH models.The rat models in the acupuncture group were given acupuncture at"Baihui"-penetrating-"Qubin"acupoints for 7 consecutive-day.Before killing the rats in each group,Bederson neurological function score was used again to evaluate the degrees of neurological function recovery of those rats.Transmission electron microscopy was used to observe the degree of mitochondrial destruction around hemorrhage foci in model rat brain,and immunohistochemistry was used to detect the expression of FTH1,a key factor of iron death,in rat brain.Results:①Acupuncture at"Baihui"-penetrating-"Qubin"acupoints could significantly improve the recovery of neurological function of ICH rats.In the acupuncture group,Bederson neurological function score was decreased significantly at 7th day compared with 1st day,with statistically significant difference(P<0.001).At 7th day,the Bederson neurological function score in the acupuncture group was decreased significantly compared with the model group,with statistically significant difference(P<0.01).②Acupuncture at"Baihui"-penetrating-"Qubin"acupoints could significantly decrease the degree of mitochondrial destruction around hemorrhage foci in the brain tissue of model rats.③Acupuncture at"Baihui"-penetrating-"Qubin"acupoints could significantly downregulate the expression of FTH1 in the brain tissue of ICH rats.The FTH1 expression in the acupuncture group was obveriously lower than that in the model group at 7th day,with statistically significant difference(P<0.001).Conclusions:Acupuncture at"Baihui"-penetrating-"Qubin"acupoints can significantly promote the recovery of neurological function in ICH rats,which may be related to the inhibition of FTH1 expression in brain tissue,thereby inhibiting mitochondrial destruction in nerve cells.关键词
