基于网络药理学探讨姜黄素干预早发性卵巢功能不全作用机制
The action mechanism of curcumin in the intervention of premature ovarian insufficiency based on network pharmacology

Abstract
Objective:To explore the potential target and action mechanism of curcumin on premature ovarian insufficiency(POI)by network pharmacology and in vitro experiments.Methods:The target of curcumin was screened by TCMSP and DrugBank database.SwissTarget Prediction database was used to predict the drug target.The disease targets of POI were obtained from GeneCards,DisGeNET and OMIM databases.The Venn diagram of drug-disease common targets was drawn.PPI network was constructed by STRING online platform and visualized by Cytoscape 3.9.1 software to determine the potential therapeutic targets of curcumin against POI.GO function and KEGG pathway enrichment analyses were performed by DAVID database.AutoDock Tools 1.5.7 and PyMOL 3.0 software were used to verify the binding ability of curcumin to its core targets.Hydrogen peroxide(H2O2)was used to induce KGN cells to construct a cell injury model,and Western blot was used to verify the effect of curcumin on target proteins and key inflammatory pathway proteins.Results:A total of 124 drug targets and 6 097 disease targets were obtained.Taking the intersection of the two,72 targets of curcumin anti-POI were obtained,mainly involving TNF-α,AKT1,and BcL-2.The GO functional enrichment analysis showed that these targets were mainly related to inflammatory response,apoptosis process,and proliferation process.The KEGG pathway enrichment analysis showed that curcumin mainly exerted its therapeutic effect on POI through signaling pathways such as HIF-1,TNF,JAK-STAT,and PI3K-Akt signaling pathway.Molecular docking results showed that curcumin had a good binding affinity with TNF-α and BcL-2.The experimental results indicated that curcumin could reverse the expressions of TNF-α,IκBα,p-IκBα,NF-κB,and BAX/BcL-2 proteins(P<0.01,P<0.001,P<0.000 1).Conclusion:Curcumin may play a role in improving ovarian function by regulating the expressions of TNF-α/NF-κB and apoptosis-related proteins.关键词
