基于GEO数据挖掘、网络药理学与分子对接探讨天麻与防风治疗急性脑梗死的有效成分和作用机制
Exploring the effective components and action mechanisms of Tianma and Fangfeng in the treatment of acute cerebral infarction through GEO data mining plus network pharmacology and molecular docking

Abstract
Objective:To investigate the key active components,targets,and pathways of Tianma(Rhizoma Gastrodiae)and Fangfeng(Radix Saposhnikoviae)in the treatment of acute cerebral infarction through network pharmacology,molecular docking and GEO data mining methods,so as to explore the potential pharmacological basis,molecular mechanisms,as well as their common action mechanisms.Methods:The main active components and related targets of Tianma and Fangfeng were screened and collected from databases such as TCMSP,PubChem,and SwissADME.Acute cerebral infarction-related targets were retrieved through GeneCards and DisGeNET databases.The targets of acute cerebral infarction were mapped with the action targets of Tianma and Fangfeng to obtain the effective intersection of medicine-disease targets.A protein-protein interaction(PPI)network was constructed by STRING database,and the results were visualized by Cytoscape 3.8.2.GO and KEGG pathway enrichment analyses were conducted through the DAVID database to identify relevant signaling pathways.Molecular docking was performed to verify the interactions between the active components of Tianma and Fangfeng and the key targets.The GEO database GSE102541 dataset was used for clinical cohort analysis to obtain differences in mRNA expression of key targets in acute cerebral infarction patients.Results:A total of 29 active components were obtained from Tianma,with 980 corresponding targets.There were 10 active components from Fangfeng,with 509 corresponding targets.After integration and deduplication,899 acute cerebral infarction-related targets were identified,with 104 genes related to Tianma and 118 genes related to Fangfeng.A total of 73 key targets and 121 signaling pathways related to Tianma,Fangfeng,and acute cerebral infarction were identified ultimately.Molecular docking results suggested good binding activities between the active components and key targets.Through the GEO database GSE102541 dataset,it was confirmed that PPARG expression in acute cerebral infarction patients was significantly higher than that of patients in the control group(P=0.007).Conclusion:Tianma and Fangfeng may exert therapeutic effects in the treatment of acute cerebral infarction by regulating processes such as oxidative stress,inflammatory response,and platelet aggregation,providing a theoretical basis for the clinical application of the two Chinese medicines.关键词
