基于网络药理学和网络毒理学探讨黄药子对甲状腺结节及肝损伤的毒效机制研究
Exploration on the action mechanism of deleterious effects of Huangyaozi on thyroid nodules and liver injury based on network pharmacology and network toxicology

Abstract
Objective:To preliminarily explore the action mechanism of Huangyaozi(Rhizoma Dioscoreae)in treating thyroid nodule and its deleterious influence on liver injury through network pharmacology and network toxicology.Methods:The related components of Huangyaozi and their corresponding targets were collected from the TCMSP.The target gene names were retrieved from the UniProt database.The targets related to thyroid nodules and hepatotoxicity were retrieved from GeneCards and OMIM databases,which were cross-linked with medicine targets respectively to obtain the common targets of medicine targets and disease targets.The common targets were imported into the STRING database for protein interaction analysis,and a visual analysis was performed by the Cytoscape software.The R language software was used to conduct GO biological analysis and KEGG signaling pathway enrichment analysis for core targets.Results:After screening,15 chemical components and 203 targets of Huangyaozi were obtained.There were 130 intersection targets of Huangyaozi and thyroid nodules,and 180 associated pathways.There were 178 intersection targets of Huangyaozi and hepatotoxicity,and 192 associated pathways.Huangyaozi could treat thyroid nodules by regulating AGE-RAGE,IL-17,TNF and other signal pathways through targets such as AKT1,TP53,TNF,IL-6,and IL-1B.The hepatotoxicity of Huangyaozi was closely related to regulating AGE-RAGE,IL-17,TNF,and other signaling pathways through AKT1,TNF,IL-6,TP53,IL-1B and other targets.Conclusion:The therapeutic effect of Huangyaozi on thyroid nodules and its deleterious influence on liver injury involve multiple components,targets,and signaling pathways,providing new ideas for subsequent in-depth research.关键词
