Abstract
Objective:The action mechanism of Yuxingcao(Herba Houttuyniae)in the treatment of nephrotic syndrome by network pharmacology and molecular docking technology,in order to explore its molecular efficacy evidence and provide a theoretical basis for its clinical application.Methods:In the TCMSP and BATMAN databases,in combination with relevant literature,taking oral bioavailability≥30%and Drug-likeness≥0.18 as standards,76 active components(such as quercetin and hexanoic acid)were screened out,and 1 323 targets were predicted.A total of 1 012 targets related to nephrotic syndrome were collected from GeneCards and DisGeNET databases.The drug-component-target-disease network was constructed by Venny 2.1,and 205 intersecting targets and core components were obtained.The protein-protein interaction network was constructed by the STRING and Cytoscape,and key targets such as ALB and AKT1 were identified.GO function and KEGG pathway enrichment analyses were conducted by Bioconductor bioinformatics software package,revealing biological processes such as antioxidation and immune regulation,as well as signaling pathways such as AGE-RAGE.Results:It was predicted that the active components of Yuxingcao in the treatment of nephrotic syndrome included quercetin,caproic acid,thymol,etc.These components might exert therapeutic effects on nephrotic syndrome by acting on targets such as ALB,INS,AKT1,IL-6,and TNF,and through cellular biological processes such as antioxidation,antibacterial,and immune regulation.The results of molecular docking showed that piperolactam A had the strongest binding affinity with ALB and TNF,confirming at the molecular level the potential action mechanism of Yuxingcao in the treatment of nephrotic syndrome.Conclusion:Yuxingcao exhibits the characteristics and advantages of multi-component,multi-target,and multi-pathway in the treatment of nephrotic syndrome.Its active components exert therapeutic effects by regulating inflammatory factors,metabolism,oxidative stress and other pathways.The conclusion provides a theoretical basis for the clinical application of Yuxingcao in the treatment of nephrotic syndrome and also lays a foundation for the further development of anti-nephrotic syndrome medicines based on Yuxingcao.关键词
