m6A methyltransferase METTL3 activates lncRNA NEAT1/PI3K/AKT path-way to promote proliferation and invasion of gastric cancer MKN-28 cells

Abstract
Objective To investigate the expression levels of m6A methyltransferase 3(METTL3)in human gas-tric cancer(GC)tissue samples and its effects on the lncRNA nuclear paraspeckle assembly transcript 1(NEAT1)/phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)signaling pathway,cell proliferation and invasion in MKN-28 cells.Methods Quantitative Reverse Transcription-Polymerase Chain Reaction(RT-PCR)and Western blot were used to detect the expression levels of METTL3 in 6 pairs of GC tissue samples,and the effects of METTL3 on the expression levels of lncRNA NEAT1 and the activity of PI3K/AKT signaling path-way were analyzed;Methylated RNA immunoprecipitation(MeRIP)was used to detect the m6A level of lncRNA NEAT1;Cell counting Kit-8(CCK-8)and Transwell assays were used to detect the effect of METTL3 knockdown on the proliferation and invasion of MKN-28 cells.Results As compared with the adjacent normal tissues,the mRNA and protein levels of METTL3 were increased in human GC tissue samples(P=0.026;P<0.000 1).Furthermore,knockdown of METTL3 reduced the mRNA levels of lncRNA NEAT1(P=0.001 8)and its m6A levels(P=0.001 5).Knockdown of METTL3 could inhibit the proliferation activity(P=0.011)and invasion ability(P=0.000 3)of MKN-28 cells,and reduced the protein levels of p-PI3K and p-AKT(P<0.001);Overexpression of NEAT1 promoted cell proliferation(P=0.024)and invasion(P=0.004),enhanced the protein levels of p-PI3K and p-AKT(P<0.01),and reversed the anti-tumor effect caused by METTL3 knock-down(P<0.01).Conclusion METTL3-mediated m6A modification of lncRNA NEAT1 activated PI3K/AKT signaling and promoted the proliferation and invasion of gastric cancer MKN-28 cells.关键词
