冰草高蛋白含量相关EST-SSR标记开发与验证
Development and validation of EST-SSR markers associated with high protein content in wheatgrass

Abstract
Crude protein content is one of the important nutritional quality traits for wheatgrass.To obtain the EST-SSR markers which are closely related to crude protein content,we used the F2 populations of a tetraploid crested wheatgrass which was generated by doubling the hybrid genome of Agropyron mongolicum and A.cristatum,and then used bulked segregant analysis(BAS)technology to develop EST-SSR marks.A total of 42 pairs of EST-SSR primers were finally amplified by polymerase chain reaction(PCR)to develop EST-SSR markers related with high protein content in wheatgrass.The F2 segregation individuals were used for validation and correlation analysis of marker sites.The results showed that the EST-SSR marker SSR-BC5-82 was closely correlated with crude protein content of wheatgrass.This EST-SSR marker can significantly and accurately mark crude protein content,and will be important in breeding new wheatgrass germplasm with high protein content and in enriching the molecular marker types of wheatgrass.关键词
