本研究基于网络药理学和分子对接技术,探讨香叶木素(Diosmetin)作为饲料添加剂对比格犬应激反应的调节作用及潜在机制.通过网络药理学分析,筛选出香叶木素与应激相关的 22 个交集靶点(占总靶点 3.8%),其中EGFR、PTGS2、ESR1 和AKT1 被鉴定为核心靶点.GO富集分析表明,香叶木素可能通过调控细胞对有机氮化合物的反应、跨膜蛋白酪氨酸激酶活性等生物学功能发挥抗应激作用;KEGG通路分析揭示其作用机制涉及内分泌抵抗、VEGF信号通路及ERBB信号通路等.分子对接结果显示,香叶木素与核心靶点PTGS2(结合能-6.95 kcal/mol)、ESR1(-6.82 kcal/mol)和EGFR(-6.33 kcal/mol)具有较强结合能力,验证了其多靶点调控潜力.本研究首次从网络药理学角度阐明了香叶木素抗应激的分子机制,为开发犬类抗应激饲料添加剂提供了理论依据,后续需通过体内试验进一步验证其实际应用效果.

This study investigated the regulatory effects and underlying mechanisms of diosmetin as a dietary supplement on stress re-sponses in beagles using network pharmacology and molecular docking approaches.Network pharmacology analysis identified 22 over-lapping targets(3.8%of total targets)between diosmetin and stress-related genes,with EGFR,PTGS2,ESR1,and AKT1 identified as core targets.Gene Ontology(GO)enrichment analysis suggested that diosmetin may exert anti-stress effects by modulating biological processes such as cellular response to organic nitrogen compounds and transmembrane receptor protein tyrosine kinase activity.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis revealed its potential mechanisms involving endocrine resistance,VEGF signaling pathway,and ERBB signaling pathway.Molecular docking results demonstrated strong binding affinities between diosmetin and key targets,including PTGS2(binding energy:-6.95 kcal/mol),ESR1(-6.82 kcal/mol),and EGFR(-6.33 kcal/mol),supporting its multi-target regulatory potential.This study is the first to elucidate the molecular mechanisms of diosmetin's anti-stress effects from a computational biology perspective,providing a theoretical foundation for developing canine anti-stress feed additives.Further in vivo studies are required to validate its practical efficacy.
