Development of rapid propagation system of Hosta plantaginea by open culture in vitro

Abstract
[Objective]Under the condition of adding antibacterial agents to the culture medium,the optimal ratio of anti-bacterial agents suitable for open tissue culture and the appropriate exogenous hormone ratio for different tissue culture stages were screened,thereby establishing a rapid propagation system for open tissue culture of Hosta plantaginea.[Method]Using the Hosta'Slivery Edger'as the experimental material,open tissue culture and rapid propagation were conducted by combining four antibacterial agent formulations(including a control)with three medium compositions.At the initial culture stage,bud emergence,browning,and contamination were evaluated for each treatment.During the subcul-ture stage,proliferation rate,vitrification,and contamination were assessed.In the rooting stage,root formation and contami-nation were analyzed.[Result]At the initial culture stage,the A3B2 combination(with the bacteriostatic agent ratio of 10 mg/L potassium sorbate,10 mg/L mancozeb,and 0.01%sodium hypochlorite,and the medium being MS+2 mg/L 6-BA+0.2 mg/L NAA)had a high germination rate,low contamination rate and browning rate.During the subculture stage,the C4D2 combination(with the bacteriostatic agent ratio of 10 mg/L potassium sorbate,40 mg/L mancozeb,and 0.01%so-dium hypochlorite,and the medium being MS+3 mg/L 6-BA+0.1 mg/L NAA)had the highest proliferation coefficient,and the lowest vitrification rate and contamination rate.In the rooting stage,the E3F2 combination(with a bacteriostatic agent ratio of 10 mg/L potassium sorbate,20 mg/L mancozeb,and 0.015%sodium hypochlorite,and a medium of 1/2 MS+0.3 mg/L NAA)produced the most roots.[Conclusion]Through the screening of antibacterial agents and culture media in open tissue culture,optimal combinations of antibacterial agents and ratios of exogenous hormones in the culture medium were identified for the initial,subculture,and rooting stages of Hosta'Slivery Edger',providing both theoretical and techni-cal support for the rapid propagation of Hosta seedlings.关键词
