基于生物信息学分析儿童注意缺陷多动障碍关键基因及中药预测
Bioinformatics-based Analysis of Key Genes and Chinese Medicinal Herb Prediction for Childhood Attention Deficit Hyperactivity Disorder

Abstract
Objective:To explore the key genes,mechanisms of action and potential targeting of traditional Chinese medicine for the inci-dence of attention deficit hyperactivity disorder(ADHD)in children.Methods:The gene chip GSE205129 were obtained from the GEO database,the data quality was evaluated and differentially expressed genes(DEGs)were screened by using the R language"limma"and other extension packages,and then the differential genes were enriched by the R language"clusterProfiler"and other expansion packages to perform GO function enrichment analysis and KEGG pathway enrichment analysis,and then STRING and Cytoscape were used The software plotted the protein interaction network,selected key genes and plotted ROC curves to evaluate the diagnostic value of key genes for ADHD,and finally predicted the potential TCM for the treatment of ADHD through the SymMap database.Results:A total of 1 000 ADHD-related DEGs were found,and the enrichment analysis showed that these DEGs were mainly involved in protein syn-thesis,inflammation and infection-related pathways,and 9 key genes were screened out,all of which had high diagnostic value.It was predicted that 24 Chinese medicines such as Ruiren(Nux prinsepiae),Tufuling(China root),and Fenbuxie(Mealy yam)are mainly mild,Cold or Warm in property.The medicinal taste is mainly bitter,sweet or spicy;the liver meridian is the most in meridian tropism;They are used as Qi regulators as well as blood-activating and stasis-reducing drugs were used most frequently.Conclusion:The oc-currence and development of ADHD in children involves the joint action of multiple genes and multiple pathways,among which nine dif-ferential genes,including DHX15,ACTB,HNRNPA2B1,PABPC1,MTREX,SRSF2,XPO1,UTP3,and MTOR,may be key targets for the treatment of ADHD in children.The 24 flavors of traditional Chinese medicine led by Ruiren have high research value for the treatment of ADHD in children.关键词
