Abstract

To identify the characteristics and biocontrol potential of the endophytic strain TA-1.3 of Acalypha australis,the taxonomic status and biological characteristics of the strain TA-1.3 was determined through morphological observation,ITS(Internal Transcribed Spacer)identification technology and adaptability of the strain to environmental factors.The inhibitory effects of the endophytic strain on Rhizoctonia solani AG1-IA,R.solani AG5,Sclerotinia sclerotiorum(Lib)de Bary,S.minor Jagger and Ma-gnaporthe oryzea were evaluated using the plate confrontation method and bacteriostatic secondary metabolites.Morphological observation shows that the colony of strain TA-1.3 emits coconut fragrance,the conidia pile is in concentric circular pattern,and the conidia stem is in pyramid shape,which has the typical morphological characteristics of Trichoderma.Through ITS identification and sequence alignment,the endophytic strain TA-1.3 was identified as Trichoderma harzianum.T.harzianum TA-1.3 had the fastest growth rate on media with starch as carbon source and yeast extract as nitrogen source,respectively.It was suitable for strain TA-1.3 cultivation in a dark environment,with an optimal pH of 9 and an optimal culture temperature of 25℃.The results of the confrontation test showd that the antagonism coefficient of T.harzianum TA-1.3 against AG1-IA,AG5,S.minor and M.oryzea all reached grade I.In a metabolite bacteriostatic test,the volatile metabolites of T.harzianum TA-1.3 produced the most obvious inhibition rate on S.sclerotiorum at 82.11%.The non-volatile metabolites of T.harzianum TA-1.3 had the most significant inhibitory effect on the growth of S.minor mycelium.
