麝香保心丸调控miR-451表达抑制ApoE-/-小鼠动脉粥样硬化斑块形成
Shexiang Baoxin Pill Regulating miR-451 Expression to Inhibit ApoE-/-Atherosclerotic Plaque Formation in Mice

Abstract
Objective:To observe the effect of Shexiang Baoxin Pill on atherosclerosis in ApoE-/-mice,and to explore the regulatory mechanism.Methods:Twenty-four male ApoE-/-mice were randomly divided into model group(AS group),determinable group(Ro-suvastatin group,1.52 mg·kg-1),Shexiang Baoxin Pill low-dose group(SXBXP-L group,4.55 mg·kg-1)and Shexiang Baoxin Pill high-dose group(SXBXP-H group,9.10 mg·kg-1),with 6 mice in each group and fed with high-fat diet.In addition,6 C57BL/6J mice were given as blank group(CON group)and given gavage for 12 weeks,once a day.HE,oil red O and Masson staining were used to detect the plaque area,lipid area and collagen fiber content of mice in the aortic sinus.The serum levels of total cholesterol(TC),triglyceride(TG)and low-density lipoprotein cholesterol(LDL-C)were detected by colorimetric method.qRT-PCR was used to detect the mRNA levels of aortic miR-451 in mice,and Western blot was used to detect the expression levels of α-smooth muscle actin(α-SMA)and proliferating cell nuclear antigen(PCNA)proteins in mice.Results:Compared with the CON group,the body weight of the mice in the AS group increased(P<0.05).Compared with the AS group,the body weight of the mice in the Rosuv-astatin group and SXBXP-H group decreased(P<0.05).Compared with the CON group,the aortic sinus plaque area and lipid area of the mice in the AS group increased(P<0.05).Compared with the AS group,the aortic sinus plaque area and lipid area of the mice in the rosuvastatin group,SXBXP-L group,and SXBXP-H group were reduced(P<0.05).Compared with the CON group,the ser-um levels of TC,TG and LDL-C in the AS group were increased(P<0.05).Compared with the AS group,the serum levels of TC,TG and LDL-C in the rosuvastatin group,SXBXP-L group and SXBXP-H group were decreased(P<0.05).Compared with the CON group,the aortic miR-451 mRNA level of mice in the AS group was decreased(P<0.05).Compared with the AS group,the mice in the SXBXP-H group had an increased level of aortic miR-451(P<0.05).Compared with the CON group,the expression level of aortic α-SMA protein in the AS group was decreased,and the expression level of PCNA protein was increased(P<0.05).Compared with the AS group,the expression level of aortic α-SMA protein in the SXBXP-L group and SXBXP-H group was increased,and the expression level of PCNA protein decreased(P<0.05).Conclusion:Shexiang Baoxin Pill can inhibit atherosclerotic plaque formation and lipid accumulation in ApoE-/-mice,and its mechanism may be related to the regulation of miR-451 expression.关键词
