Screening Serum Biomarkers of Vitamin E Deficiency in Dairy Cows Based on Targeted Metabolomics

摘要

Abstract

Targeted metabolomics was used to characterize serum fatty acid and amino acid profiles of dairy cows with Vitamin E deficiency(VED)in early lactation to obtain effective serum biomarkers for the identification of VED.At 21 days postpartum,ultraperformance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)technology was used to target and quantify the changes in serum fatty acids and amino acids in cows from the VED group(α-tocopherol<3 μg·mL-1,n=10)and the normal group(α-tocopherol≥4 μg·mL-1,n=10).Compared with the normal group,8 metabolites were up-regulated and 6 metabolites were down-regulated in the serum of cows in the VED group.These differential metabolites were primarily enriched in eight metabolic pathways associated with amino acid,lipid,carbohydrate,and cofactor and vitamin metabolism.Furthermore,the diagnostic thresholds for VED in dairy cows were serum undecanoic acid concentration<0.23 μmol·L-1 and L-citrulline concentration<60.54 μmol·L-1.In conclusion,the combined index of serum undecanoic acid and L-citrulline may be effective biomarkers for the diagnosis of VED in early lactation dairy cows.It provides a new method for the clinical diagnosis of VED in dairy cows.
