猪流行性腹泻病毒感染PK15细胞的转录组学分析
Transcriptomic Analysis of PK15 Cells Infected by Porcine Epidemic Diarrhea Virus

Abstract
Porcine epidemic diarrhea virus infection can cause severe diarrhea,vomiting and dehydration in newborn piglets,with an average mortality rate of up to 80%,which brings huge economic losses and harms to the pig industry.In order to explore how innate immunity affected PEDV replication,differentially expressed gene of PEDV CV777 MOI=1 infected PK15 cells for 24 h and 48 h,respectively.GO functional analysis and KEGG pathway enrichment analysis were conducted,and some DEGs were selected for RT-qPCR validation.Transcriptomic analysis results showed that,compared with normal uninfected PK15 cells,there were 1 798 differential genes post 24 h of PEDV infection,the results of GO molecular function showed that it was mainly related to biological process regulation,response to stimuli,immune response,etc.KEGG enrichment showed that DEGs was mainly involved in cytokine and factor receptor binding pathway,PI3K-Akt signaling pathway,chemokine signaling pathway,etc.There were 3 970 differential genes post 48 h of PEDV infection,the results of GO molecular function demonstrated that DEGs was mainly related to signal transduction,response to stimulation,immune response and inflammatory response.KEGG enrichment showed that DEGs was mainly involved in the PI3K-Akt signaling pathway and MAPK signaling pathway.Further screening and statistics of PK-p24 and PK-p48 DEGs showed that there were 773 common differential genes,there are three differential genes(CCL8,PRKN,FGL2),which are enriched in four biological functions.innate immune response,response to stimulation,inflammatory response,and host replication of viral genome.RT-qPCR was used to verify the three common differential genes,and the results were basically consistent with the transcriptomic results.It is suggested that PK15 cells infected with PEDV CV777 MOI=1 might inhibit the replication of the virus by regulating the changes in the transcription levels of CCL8,PRKN and FGL2,thus enhancing the innate immune response.关键词
