钝顶螺旋藻藻蓝蛋白抗氧化酶解产物的制备及活性研究
Study on Preparation and Activity of Antioxidant Enzymatic Hydrolysates of Phycocyanin from Spirulina platensis

Abstract
To obtain the highly efficient antioxidant enzymatic hydrolysate of phycocyanin from Spirulina platen-sis,phycocyanin was used as the raw material.The DPPH·scavenging rate was taken as the index.Based on the screening of the enzymes for enzymatic hydrolysis,the single factor experiment combined with the Box-Behnken central composite response surface method was used to establish the optimal enzymatic hydrolysis process.The enzy-matic hydrolysate prepared under the optimal process was evaluated for antioxidant activity and then preliminarily purified.The results showed that the optimal enzyme for enzymatic hydrolysis was alkaline protease.The optimal en-zymatic hydrolysis process parameters for the highly efficient antioxidant enzymatic hydrolysate were as follows:en-zyme addition amount of 10 000 U/g,enzymatic hydrolysis time of 56 min,enzymatic hydrolysis temperature of 51.5℃,and enzymatic hydrolysis pH of 10.2.Under this optimal process,when the mass concentration of the enzy-matic hydrolysate was 20.00 mg/mL,the DPPH·scavenging rate was 30.33%;when the mass concentration was 7.50 mg/mL,the·OH scavenging rate was 90.86%;when the mass concentration was 0.12 mg/mL,the ABTS+·scav-enging rate was 83.49%;when the mass concentration was 10.91 mg/mL,the·O2ˉ scavenging rate was 55.60%.Af-ter purification with 80%ethanol,the DPPH·and ABTS+·scavenging rates of the purified enzymatic hydrolysate in-creased by 2.3 times and 3.8 times respectively compared to those before purification.The difference in scavenging rates against two types of radicals demonstrated statistical significance(P<0.05)before and after purification of the enzymatic hydrolysates.The quadratic model equation developed in this study demonstrated high reliability(pre-dicted R2=0.905)and can be used to predict the optimal enzymolysis process for obtaining highly efficient antioxi-dant enzymatic hydrolysates,providing a data basis for the preparation of antioxidant peptides from S.platensis by enzymatic hydrolysis and the development of anti-aging special dietary foods.关键词
