补肺汤对COPD小鼠肺组织细胞凋亡及炎症因子的影响
Effect of Bufei Decoction(补肺汤)on apoptosis and inflammatory factors in lung tissues of mice with chronic obstructive pulmonary disease

Abstract
Objective To investigate the effect of Bufei Decoction(补肺汤,BFD)on apoptosis and inflammatory factors in the lung tis-sues of mice with chronic obstructive pulmonary disease(COPD).Methods Mice were divided into four groups:the normal control group,COPD model group,BFD group,and the azithromycin group.A COPD mouse model was established by passive cigarette smoke(CS)exposure and intratracheal lipopolysaccharide(LPS)instillation.The forced expiratory volume in 0.3 seconds to forced vital capacity ratio(FEV0.3/FVC)was measured.Pathological changes in lung tissue were assessed by HE staining.Levels of IL-1β,TNF-α,and IL-6 in bronchoalveolar lavage fluid(BALF)were determined by ELISA.The apoptosis rate of lung tissue cells was evaluated by TUNEL staining.Expression levels of endoplasmic reticulum stress-related proteins GRP78,Bax,Caspase-3,and Bcl-2 were detected by Western blot(WB).Results After 1 month of modeling,lung function in the model group decreased significantly compared with the normal control group.Compared with the normal control group,the model group showed a decreased FEV0.3/FVC,pathological lesions in lung tissues observed by HE staining,elevated levels of IL-6,IL-1β,and TNF-α in BALF(P<0.05),an increased apoptosis rate of lung tissue cells(P<0.05),upregulated expression of endoplasmic reticulum stress-related proteins GRP78,Bax,and Caspase-3(P<0.05),and downregulated expression of the anti-apoptotic protein Bcl-2.Compared with the model group,both the BFD group and the azithromycin group exhibited an increased FEV0.3/FVC(P<0.05),ameliorated pathological lung tissue damage observed by HE staining,decreased levels of IL-6,IL-1β,and TNF-α in BALF,and a reduced apoptosis rate of lung tissue cells.Moreover,these treatments downregulated the expression of GRP78,Bax,and Caspase-3(P<0.05)and upregulated the expression of Bcl-2(P<0.05)compared with the model group.Conclusion BFD can inhibit apoptosis in lung tissues,improve lung function,and attenuate inflammation in COPD mice.This protective effect may be associated with modulation of the endoplasmic reticulum stress-related pro-tein GRP78.关键词
