参芪补中方通过Nrf2/SLC7A11/GPX4信号通路调控铁死亡改善COPD大鼠炎症的研究
Study on alleviation of inflammation in COPD rats by regulating ferroptosis through the Nrf2/SLC7A11/GPX4 signaling pathway by Shenqi Buzhong Formula(参芪补中方)

Abstract
Objective To investigate whether Shenqi Buzhong Formula(参芪补中方,SQBZ)reduces inflammation in COPD rats by regulating ferroptosis through the Nrf2/SLC7A11/GPX4 signaling pathway.Methods COPD rat models were established by cigarette smoke exposure combined with intratracheal instillation of lipopolysaccharide(LPS).Rats were divided into the model group,low/medium/high-dose SQBZ groups,and the blank control group.The MDA level was detected 24h after the last administration.Another 18 COPD rats were established and randomly divided into the model group,SQBZ group,SQBZ+ML385 group,and a blank control group.The general condition of rats was observed.Pathological changes in lung tissues were assessed by HE staining,and mitochon-drial morphology was examined using electron microscopy.The expression levels of SOD,GSH-Px,GSH and MDA were detected,and the levels of inflammatory factors IL-1β,and TNF-α were quantified by ELISA.PCR was used to detect the mRNA expression levels of Nrf2,SLC7A11 and GPX4 in lung tissues,and Western blot(WB)was used to detect the protein expressions of Nrf2,SLC7A11 and GPX4 in lung tissues.Results Compared with the blank control group,the model group showed severe inflammatory cell infiltration in lung tissues,disrupted mitochondrial cristae,and even mitochondrial structural loss.Body weight,SOD and GSH-Px activities,and protein expression of Nrf2,SLC7A11,and GPX4 were significantly decreased(P<0.05),and the expression of inflammatory factors IL-1β,and TNF-α and lipid peroxidation product MDA was elevated in lung tissues(P<0.05).Compared with the SQBZ group,the SQBZF+ML385 group attenuated the therapeutic effect of SQBZ,with heavier inflammatory cell infiltration in the lung tissues of the rats,lower levels of body mass,SOD,GSH-Px and protein expressions of Nrf2,SLC7A11 and GPX4(P<0.05),and elevated expres-sions of IL-1β,and TNF-α and MDA in lung tissues(P<0.05).Conclusion SQBZ may alleviate COPD by inhibiting ferroptosis through activation of the Nrf2/SLC7A11/GPX4 signaling pathway,thereby reducing inflammation.关键词
