摘要

Abstract

Objective To evaluate the therapeutic effects and mechanisms of Qiangzhi Formula(强志方,QZF)in a rat model of tic disorder(TD)combined with attention deficit hyperactivity disorder(ADHD),providing experimental data and theoretical basis for the prevention and treatment of TD combined with ADHD.Methods Six Sprague-Dawley(SD)rats were designated as the blank control group,while 30 spontaneously hypertensive rats(SHR)were randomly divided into the sham surgery group,model group,and low/medium/high-dose QZF groups.Behavioral changes were assessed using stereotypy scores,locomotor scores,open field test(OFT),elevated plus maze test(EPMT),and Morris water maze test(MWMT).Western blot(WB)and RT-PCR were used to detect the expression levels of calcium/calmodulin-dependent protein kinase Ⅱ(CaMK Ⅱ),tyrosine hydroxylase(TH),and dopamine(DA)-related proteins and mRNA.Results Stereotypy and locomotor scores were significantly increased in the model group but decreased notably after QZF treatment,with the most significant improvement observed in the high-dose group.Behavioral tests showed significant improvements in locomotor indicators in OFT and EPMT in the high-dose group,as well as shortened escape latency and increased target quadrant-related indicators in the MWMT.Western blot(WB)and RT-PCR revealed that compared to the model group,the high-dose QZF group exhibited significantly reduced expressions of P-TH,P-CaMK Ⅱ,and DA-related proteins,alongside decreased mRNA expression of CaMK Ⅱ,TH,and DA.Conclusion QZF significantly alleviates tic,hyperactivity,and attention deficit symptoms in the TD combined with ADHD rat model.Its mechanism may be associated with the regulation of DA-related pathways by CaMK Ⅱ.
