补血再生糖浆调节骨髓巨噬细胞M1/M2极化治疗再生障碍性贫血的作用机制
Mechanism of Buxue Zaisheng Syrup(补血再生糖浆)in ameliorating aplastic anemia in mice by regulating M1/M2 polarization of bone marrow macrophages

Abstract
Objective To investigate the effects and mechanism of Buxue Zaisheng Syrup(补血再生糖浆,BXZSS)on aplastic anemia(AA)in mice.Methods Fifty female BALB/c mice were randomly divided into the blank group,model group,high/low-dose BXZSS groups,and the cyclosporine A group.Except for the blank group,the other groups were intraperitoneally injected with cyclophospha-mide(25 mg/kg)for 14 consecutive days to establish the AA model.After modeling,the corresponding drugs were administered to each group for 14 days.The general condition and body weight of the mice were observed and recorded.The numbers of white blood cells(WBC),red blood cells(RBC),platelets(PLT),and hemoglobin(HGB)in the peripheral blood of the mice were measured.The number of bone marrow mononuclear cells was observed by Wright-Giemsa staining.The serum levels of IFN-γ,TNF-α,IL-4,and TGF-β were detected by ELISA.The plasma levels of granulocyte colony-stimulating factor(G-CSF),granulocyte-macrophage colony-stimulating factor(GM-CSF),erythropoietin(EPO),and thrombopoietin(TPO)were detected by ELISA.The expression levels of CD68 and CD206 on bone marrow cells were detected by flow cytometry.Results Compared with the blank group,the model group showed significantly reduced average body weight,peripheral blood cell counts(WBC,RBC,PLT)and hemoglobin(HGB)levels,number of CD68+bone marrow macrophages,and serum levels of IFN-γ and TGF-β(P<0.01).The model group also exhibited sig-nificantly increased plasma levels of G-CSF,GM-CSF,EPO,and TPO,number of mature bone marrow macrophages,serum levels of IFN-γ and TNF-α,number of CD206+macrophages,and CD68+/CD206+macrophage ratio(P<0.01).Compared with the model group,mice in the high-dose BXZSS group and the cyclosporine A group showed significant improvements in the aforementioned indica-tors(P<0.05 or P<0.01).Conclusions BXZSS ameliorates bone marrow hematopoietic function in AA mice,which may be related to its regulation of M1/M2 polarization in bone marrow macrophages.关键词
