Effects of Tongxie Yaofang(痛泻要方)on intestinal microbiota and short-chain fatty acids in DSS-induced mice with ulcerative colitis

Abstract
Objective To investigate the effects of Tongxie Yaofang(痛泻要方,TXYF)on intestinal microbiota and the metabolite short-chain fatty acids(SCFAs)in dextran sulfate sodium(DSS)-induced ulcerative colitis(UC)mice.Methods 27 SPF-grade male Balb/c mice were randomly divided into groups.The model and treatment groups received 5%(w/v)DSS in drinking water to induce UC.The treatment group was administered TXYF(11.70 g/kg)via oral gavage for 7 days.Post-intervention,serum levels of IL-10,IL-6,TNF-α,IL-1β,and TGF-β were measured using ELISA.Ileal contents were subjected to 16S rRNA gene sequencing and SCFAs quantification.Results TXYF promoted mucosal structural recovery,alleviated inflammatory infiltration,increased serum IL-10 levels,and reduced serum IL-6 and TNF-α levels(P<0.05).Sequencing identified 407 shared operational taxonomic units(OTUs).TXYF intervention elevated the Chao1 and Faith's phylogenetic diversity(PD)indexes,significantly altered Beta diversity(P<0.01),increased the relative abundance of the Verrucomicrobia and TM7 phyla,and the Firmicutes/Bacteroidetes ratio(P<0.05).Further-more,18 bacterial taxa showed differential abundance compared to controls.At the genus level,serum TGF-β1 levels were significantly correlated with the relative abundance of Desulfovibrio and Sulfuricurvum(P<0.01),while serum IL-6 levels were significantly corre-lated with the relative abundance of Subdoligranulum and Coprococcus(P<0.01).At the phylum level,serum TNF-α levels were cor-related with the relative abundance of Verrucomicrobia(P<0.05),and serum IL-1β levels were correlated with the relative abundance of TM7,TM6,and OD1(P<0.05).SCFAs quantification indicated TXYF elevated acetic and butyric acid concentrations while reduc-ing propionic acid(P<0.05).Conclusion TXYF alleviated mucosal barrier damage in colitis models by reducing pro-inflammatory cytokines(IL-6,TNF-α),elevating levels of acetic acid and butyric acid,and lowering the level of propionic acid,highlighting its association with modulation of gut microbiota and SCFAs metabolism.关键词
