基于数据挖掘和网络药理学探讨国医大师朱良春治疗痛风的用药规律及作用机制
Exploration of medication patterns and mechanisms for gout treatment by Master Zhu Liangchun through data mining and network pharmacology

Abstract
Objective:To summarize medication patterns and explore mechanisms of National Master of Chinese Medi-cine Zhu Liangchun's gout treatments based on Gout(Turbid Stasis Impediment)Diagnosis,Treatment and Rehabilitation Manual.Methods:Medical records were analyzed to determine herb frequencies,properties,flavors,and meridian tro-pism;core herb combinations were identified through association rule mining and hierarchical clustering;network pharma-cology and molecular docking techniques elucidated therapeutic mechanisms.Results:92 prescriptions containing 227 herbs revealed herbs predominantly exhibiting neutral,warm,or cold properties;sweet,bitter,or pungent flavors;and liver,spleen,or stomach meridian tropism.Core combination:Tufuling,Weilingxian,Zexie,Yiyiren,Bixie.TCMSP identified 105 active components targeting 190 herb targets,intersecting with 100 of 2,321 gout targets from GeneCards/OMIM/Drug-Bank.PPI networks identified 10 core targets:TNF,IL-6,MMP-9,Akt1,BCL2,IL-1β,PTGS2,IFNγ,CXCL8,CCL2,GO/KEGG enrichment indicated regulation via PI3K-Akt and NF-kB pathways,linked to lipid metabolism,athero-sclerosis,and diabetic complications.Molecular docking confirmed stable binding between quercetin/kaempferol/naringenin and TNF/IL-6/MMP-9.Conclusion:Professor Zhu treated gout primarily through heat-clearing,wind-damp dispelling,impediment-dredging,diuresis-promoting,and stasis-transforming methods;core components(e.g.,quercetin,stigmas-terol,β-sitosterol)modulate TNF/Akt/IL-6 targets via PI3K-Akt and NF-κB pathways.关键词
