Abstract

Objective To analyze the effect of DLD gene on the cell model of steroid-induced femoral head necrosis(SNFH)using bone marrow mesenchymal stem cells(BMSCs)via the PI3K/AKT signaling pathway.Methods BMSCs were cultured and divided into four groups:BMSCs,Model,Model+si-NC,and Model+si-DLD.The expression of DLD was analyzed using polymerase chain reaction(PCR)and Western Blot(WB).Cell proliferation and apoptosis were assessed using CCK-8 and TUNEL assays,respectively.Oxidative stress levels were determined by measuring reactive oxygen species(ROS).PCR and WB were also employed to evaluate markers of osteogenesis(RUNX2),adipogenesis(PPAR-γ),apoptosis(Bax/Bcl-2),angiogenesis(VEGF),oxidative stress(HO-1),and PI3K/AKT pathway activity.Bone formation and adipogenic differentiation were analyzed using Alizarin Red and Oil Red O staining.Results Overexpression of DLD decreased in the levels of p-PI3K and p-AKT,thereby inhibiting the PI3K/AKT pathway.This results in reduced calcium deposition,increased adipocyte formation,elevated apoptosis,decreased angiogenesis,enhanced oxidative stress,and impaired cell proliferation.Conclusion The DLD gene inhibits the activation of the PI3K/AKT signaling pathway,thereby suppressing osteogenic differentiation and promoting adipogenic differentiation in the BMSCs cell model of SNFH.
