摘要

Abstract

The unique aquatic environment oflotus(Nelumbo nucifera)brings numerous biological and abiotic stresses to its growth and development.Proanthocyanidins serve as a crucial defense mech-anism for plants against these stresses.However,the molecular mechanism underlying their synthe-sis in lotus remains unclear.We screened and cloned NnMYB6-like gene related to proanthocyanidin synthesis in lotus,performed bioinformatics analysis,and conducted transient overexpression in'Taikonglian'to analyze its function in the proanthocyanidin biosynthesis pathway by RNA-Seq.The results showed that transient overexpression of NnMYB6-like gene in'Taikonglian'lotus leaves sig-nificantly up-regulated the expression of key genes involved in the anthocyanin synthesis pathway,in-cluding NnANS,NnF3H and NnLAR,compared to the wild-type.Notably,NnLAR played a cruci-al role in proanthocyanidin metabolism.Moreover,the proanthocyanidin content was substantially in-creased in lotus leaves with transient overexpression.The transiently overexpressed lotus displayed a significantly higher proanthocyanidin content compared to the wild-type,suggesting that the NnMYB6-like gene may exert positive regulation on the proanthocyanidin synthesis pathway in lotus.The results provide a theoretical foundation for the exploration of the molecular mechanism of proan-thocyanidin synthesis in lotus.
