Abstract

To obtain U87-Exos@RES,U87 cell exosomes(U87-Exos)were extracted from the conditioned medium of U87 cells by ultracentrifugation,and resveratrol(RES)was encapsulated with them by ultrasonication.The mor-phology,particle size distribution,and surface marker proteins of U87-Exos and U87-Exos@RES were character-ized by transmission electron microscopy(TEM),nanoparticle tracking analysis(NTA),and protein blotting(West-ern Blot),confirming the successful construction of U87-Exos@RES.The homologous tumor targeting of U87-Exos was investigated using PKH26 to label U87-Exos.Furthermore,the therapeutic efficacy and mechanism of action of U87-Exos@RES were investigated.Cellular uptake experiments indicated that U87-Exos had homologous tumor tar-geting and promising application as a targeted drug delivery vector.In vitro experiments showed that U87-Exos@RES was highly cytotoxic and inhibited tumor cell proliferation by down-regulating the PI3K/AKT signaling path-way.In conclusion,U87-Exos@RES represents a novel drug delivery system that could privide a new strategy for targeted therapy of glioblastoma multiforme(GBM).
