基于16S rDNA的高通量测序技术在口虾蛄食性分析中的可行性
Feasibility of 16S rDNA-Based High-Throughput Sequencing Technology in Dietary Analysis of Oratosquilla oratoria

Abstract
In October,during the period of high feeding intensity of Oratosquilla oratoria,samples were collected from Dongying near the Yellow River estuary and Longkou,located farther from the Yellow River estuary.The gas-tric contents of O.oratoria were analyzed using 16S rDNA-based high-throughput sequencing technology to evaluate its feasibility for studying the feeding habits of O.oratoria.A total of 220 valid OTUs were obtained,which were annotated across 4 phyla,6 classes,20 orders,32 families,and 45 genera.In the waters near the Yellow River es-tuary,the highest relative abundance of dietary components was O.oratoria(91.08%),followed by mollusks(3.72%),fishes(3.71%),and shrimps(1.39%).After excluding the high abundance of O.oratoria se-quences,the sequencing results better aligned with findings from gastric content observations.In the area far from the Yellow River estuary,shrimp had the highest relative abundance(41.75%),followed by fish(24.51%),de-tritus(16.12%),and O.oratoria(15.22%),which also aligned more closely with gastric content observations.The 16S rDNA-based high-throughput sequencing demonstrated strong potential for determining the trophic ecologi-cal niche of O.oratoria.However,it should be supplemented with gastric content observations to clarify food in-take pathways and prey status(e.g.,eggs,larvae,juveniles,dead individuals).Further refinement of amplifica-tion primers and rational calibration of sequencing results are needed for effective application of 16S rDNA sequen-cing in the dietary analysis of O.oratoria.关键词
