Effect of electroacupuncture on chronic inflammation in mice with simple obesity via regulating the interaction of HIF-1α and TLR4/NF-κB

摘要

Abstract

Objective It is to observe the effects of electroacupuncture(EA)on the interactions of hypoxia-inducible factor-1α(HIF-1α)and Toll-like receptor 4(TLR4)/nuclear factor-κB(NF-κB)in peritesticular fat of mice with simple obese,and to explore the mechanism of EA in the treatment of obesity.Methods Fifty C57BL/6 mice were randomly taken,in which 12 mice were randomly selected as normal group,and the remaining mice were fed with high-fat diet for 8 weeks to establish models of simple obesity.The successfully modeled mice were randomly divided into 3 groups:model group,EA group and lipopolysaccharide(LPS)group,with 12 mice in each group.The mice of the EA group were treated with EA at acupoints including Zhongwan(CV12),Guanyuan(CV4),Zusanli(ST36)and Fenglong(ST40)once daily for 4 weeks,the mice of the LPS group were treated with LPS 0.1 mg/kg by intraperitoneal injection every other day for 4 weeks,the mice of the control and model groups were only grabbed and fixed without additional interventions.The Lee's index of mice in each group was calculated before and after treatment,the structure of peritesticular adipose tissue cells was observed by HE staining,the expression of HIF-1α and TLR4-labeled adipocytes in peritesticular adipose tissue was ob-served by immunofluorescence staining,the protein expressions of HIF-1α,TLR4,NF-κB and MyD88 in peritesticular adi-pose tissue were detected by Western blot method.Results After 4 weeks of treatment,the Lee's index of mice in the mod-el group was significantly higher than that in the normal group(P<0.05),while the Lee's index in the EA group was sig-nificantly lower than those in the model group and LPS group(both P<0.05).HE staining showed that the morphology of peritesticular adipocytes of mice in all modeled groups were disorganized with enlarged volume,the adipocyte volume of LPS group was larger than that of the model group,while the adipocyte morphology and volume in the EA group were slightly im-proved compared with those in the model and LPS group.Compared with the normal group,the number of HIF-1α and TLR4 labeled cells in peritesticular adipose tissue of each modeled group was significantly increased,and the number of la-beled cells was the largest in the LPS group,while the labeled cells in the EA group was fewer than those in both the model group and LPS group.The relative protein expressions of HIF-1α,TLR4,NF-κB and MyD88 in peritesticular adipose tis-sue were significantly higher in the model group and LPS group than those in the normal group(all P<0.05),while the relative protein expressions of HIF-1α,TLR4,NF-κB and MyD88 in the EA group were significantly lower than those in the model group and LPS group(all P<0.05),the relative protein expressions of TLR4 and MyD88 in the LPS group were significantly higher than those in the model group(both P<0.05).Conclusion Electroacupuncture can reduce body fat content in mice with simple obesity,the effects may be achieved by regulating the interaction of HIF-1α and TLR4/NF-κB to inhibit chronic inflammatory responses in adipose tissue during obesity.
