金果胃康胶囊对胃癌前病变大鼠NF-κB/HIF-1α/BNIP3信号通路的影响
Effects of Jinguo Weikang Capsules on NF-κB/HIF-1α/BNIP3 signaling pathway in rats with precancerous lesions of gastric cancer

Abstract
Objective It is to explore the effect of Jinguo Weikang Capsules on gastric mucosal lesions and the nuclear factor-κB(NF-κB)/hypoxia-inducible factor-1α(HIF-1α)/BNIP3 signaling pathway in rats with gastric precancerous le-sions.Methods Seventy-five male SPF-grade Wistar rats were taken,in which 12 rats were randomly selected as blank group,the remaining rats were continuously treated for 12 weeks with 1-methyl-3-nitro-1-nitrosoguanidine(MNNG),raniti-dine,45%ethanol and an irregular feeding regimen to establish models of gastric precancerous lesion.The successfully mod-eled rats were randomly divided into model group,folic acid group,high-dose,medium-dose and low-dose groups of Jinguo Weikang Capsule,with 12 rats in each group.The high-dose,medium-dose,and low-dose groups of Jinguo Weikang Capsule were treated with Jinguo Weikang solution by gavage at doses of 1.8 g/(kg·d),0.9 g/(kg·d),and 0.45 g/(kg·d),re-spectively,the folic acid group was treated with folic acid solution by gavage at a dose of 2 mg/(kg·d),the model group and blank group were given deionized water 2 mL/d by gavage.After 8 weeks of continuous gastric lavage,tissue samples were collected.The histopathological changes in gastric mucosa were observed by HE staining,the positive protein expres-sion of BNIP3 was observed by immunohistochemistry,mRNA expressions of NF-κB p65 and HIF-1α in gastric tissue were detected by qPCR,the protein expressions of NF-κB p65,p-NF-κB p65 and HIF-1α in gastric tissue were detected by Western blot.Results The arrangement of gastric mucosal glands of rats in the model group was disorganized with multifocal atrophy,accompanied by intestinal metaplasia and dysplasia,the gastric mucosal lesions were improved in all medicated groups,and the improvements were the most significant in the high-dose group of Jinguo Weikang Capsule.Compared with the blank group,the mean optical density value of positive protein expression of BNIP3,as well as relative mRNA expres-sions of NF-κB p65 and HIF-1α,and relative protein expressions of NF-κB p65,p-NF-κB p65 and HIF-1α in gastric tis-sue of rats in the model group were significantly increased(all P<0.05).Compared with the model group,the mean opti-cal density value of positive protein expression of BNIP3 in gastric tissues of rats in the folic acid group and high-and low-dose groups of Jinguo Weikang Capsule,as well as the relative mRNA expressions of NF-κB p65 and HIF-1α,the relative protein expressions of NF-κB p65,p-NF-κB p65,HIF-1α in gastric tissues of rats in the folic acid group and the medium-and high-dose groups of Jinguo Weikang Capsules were significantly decreased(all P<0.05).Conclusion Jinguo Wei-kang Capsule can alleviate gastric mucosal inflammation and repair gastric mucosal lesions maybe via inhibiting NF-κB/HIF-1α/BNIP3 pathway to prevent and treat precancerous lesions of gastric cancer.关键词
