Abstract

Objective It is to observe the effect of decoction for tonifying kidney and unblocking collaterals on the tumor necrosis factor receptor-associated factor 6(TRAF6)/nuclear factor-κB(NF-κB)/inducible nitric oxide synthase(iNOS)signaling pathway in rats with knee osteoarthritis(KOA),and to explore its therapeutic mechanism in the treat-ment of this disease.Methods Eighty male SD rats were divided into 5 groups by a random number table,with 16 rats in each group.The rats of all groups except for the normal group were prepared to establish models of KOA by Hulth method.After 4 weeks of modeling,the low-,medium-,and high-dose groups of decoction for tonifying kidney and unblocking col-laterals were given decoction for tonifying kidney and unblocking collaterals by gavage at the doses of 1.105 mL/200 g,2.21 mL/200 g,and4.42 mL/200 g,respectively,the normal group and model group were given physiological saline by gavage at dose of 2.21 mL/200 g,all once daily,continuously administered for 2 weeks.The general conditions and X-ray imaging examination results of the rats were observed,the synovial and cartilage tissue lesions in knee joint were observed by HE staining and toluidine blue staining,the serum levels of interleukin-6(IL-6),interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)were measured by ELISA,and the mRNA and protein expressions of TRAF6,p-NF-κB p65 and iNOS in knee joint cartilage tissue were detected by qPCR and Western blot methods.Results Compared with the normal group,the rats in the model group were languid with reduced activity and poor appetite,X-ray examination revealed signifi-cant narrowing or even disappearance of the knee joint space,roughening of the tibial plateau surface,and hardening of the distal femur accompanied by osteophyte formation,with severe cartilage tissue destruction.Compared with the model group,the injuries of knee joint cartilage tissue in rats of the medium-,and high-dose groups of decoction for tonifying kidney and unblocking collaterals were alleviated.The serum levels of IL-6,IL-1β and TNF-α,as well as the relative mRNA and pro-tein expressions of TRAF6,p-NF-κB p65 and iNOS in knee cartilage tissue in rats of the model group were significantly higher than those in the normal group(all P<0.05),the serum levels of IL-6,IL-1β and TNF-α in rats of the medium-,and high-dose groups of decoction for tonifying kidney and unblocking collaterals,as well as the relative mRNA and protein expressions of TRAF6,p-NF-κB p65 in knee cartilage tissue in rats of each dose groups of decoction for tonifying kidney and unblocking collaterals,and the relative mRNA and protein expressions of iNOS in knee cartilage tissue in rats of the high-dose group of the decoction for tonifying kidney and unblocking collaterals were all significantly lower than those in the model group(all P<0.05).Conclusion Decoction for tonifying kidney and unblocking collaterals can alleviate the symp-toms,improve cartilage tissue degeneration,and reduce the serum levels of inflammatory factors in rats with of knee osteo-arthritis,and the mechanism of action may be related to regulate TRAF6/NF-κB/iNOS signaling pathway.
