Abstract

To investigate the effects of exogenous insulin-like growth factor-I(IGF-I)on apoptosis and cell cycle mechanisms of bovine mammary gland epithelial cells(BMECs)through the PI-3K/AKT/mTOR signaling pathway.This experiment used a control group and an experimental group for mutual comparison.The experimental groups were divided into three groups,namely,experimental group 1 added 25 µg/mL IGF-I to the cells,experimental group 2 added 25 µmol/L PI-3K signal specific blocker LY294002,experimental group 3 added both inhibitor LY294002 and IGF-I,and the control group was cultured alone with only the same volume of complete culture medium as the experimental group.Each group undergoes 3 parallel treatments,with each parallel being tested twice.After 24 hours of cultivation,extract the supernatant from each group and detect cell apoptosis and cell cycle.And fluorescence quantitative PCR was used to detect the expression levels of apoptosis related genes.The experimental results showed that there was no significant difference in cell apoptosis between the experimental group and the control group.However,experimental groups 1 and 2 showed a trend of reducing cell apoptosis.For the cell cycle,compared with the control group,the G1 phase cell experimental group 1 was significantly higher than the control group(P<0.01).Compared with the control group,the G2 phase cells in experimental groups 2 and 3 were significantly higher(P<0.01).This indicates that the PI-3K/AKT/mTOR signaling pathway,as one of the important components of many pathways,plays a certain regulatory role in cell apoptosis and cell cycle transformation.IGF-I,as an anti apoptotic cytokine,can exert a certain degree of inhibitory effect on cell apoptosis through the PI-3K/AKT/mTOR signaling pathway.This mechanism may be achieved through its regulation of the cell cycle.
