重症心痹方调控焦亡关键蛋白NLRP3、ASC、caspase1干预肠源性脓毒症合并心肌损伤的作用机制研究
Study on the Mechanism of Zhongzheng Xinbi Formula in Intervening Myocardial Injury in Enterogenic Sepsis by Regulating Pyroptosis Key Proteins NLRP3,ASC,and Caspase1

Abstract
Objective:To investigate the protective effects of Zhongzheng Xinbi Formula(ZZXBF)on myocardi-al injury induced by enterogenic sepsis and to explore its pharmacological mechanisms.Methods:Rats were pre-treated with ZZXBF,and the blood components of ZZXBF were identified using liquid chromatography-tandem mass spectrometry(LC-MS/MS).Before modeling,the remaining rats were randomly divided into the sham opera-tion group(only abdominal incision and suture),the model group,the positive drug group(intraperitoneal injection of methylprednisolone 8 mg/kg),and the ZZXBF group(oral gavage of ZZXBF).A rat model of sepsis-induced myocardial injury was established using the cecal ligation and puncture(CLP)method.HE staining was used to ob-serve pathological changes in cardiac tissue;an automatic biochemical analyzer was employed to measure the lev-els of CK,CK-MB,and LDH;immunohistochemistry was performed to detect the expression levels of pyroptosis key proteins NLRP3,ASC,and Cleaved-caspase1 in cardiac tissue;Western blotting was used to assess the pro-tein levels of ASC and Cleaved-caspase1 in cardiac tissue.Results:A total of 164 blood components of ZZXBF were detected.Compared with the sham operation group,the model group exhibited significant pathological damage to myocardial tissue and a significantly increased pathological score(P<0.05);levels of CK,CK-MB,and LDH were significantly increased(P<0.05);the ratios of integrated optical density(IOD)to area(AREA)for NLRP3,ASC,and Cleaved-caspase1 proteins were significantly increased(P<0.05);and the protein levels of ASC and Cleaved-caspase1 were significantly increased(P<0.05).Compared with the model group,ZZXBF intervention sig-nificantly reduced the myocardial pathological injury score and the levels of CK,CK-MB,and LDH induced by en-terogenic sepsis(all P<0.05),significantly decreased the IOD/AREA ratios of NLRP3,ASC,and Cleaved-cas-pase1 proteins(P<0.001),suppressed the expression level of the pyroptosis key protein Cleaved-caspase1(P<0.01),and downregulated the expression level of ASC protein(P>0.05).Conclusion:ZZXBF effectively amelio-rates myocardial injury in enterogenic sepsis by inhibiting the expression of key pyroptosis proteins,thereby antago-nizing pyroptosis.关键词
