补肺汤抑制PI3K/Akt/HIF-1α/VEGFA信号通路改善大鼠急性支气管炎的机制研究
Mechanism of Bufei Decoction in Alleviating Acute Bronchitis in Rats by Inhibiting the PI3K/Akt/HIF-1α/VEGFA Signaling Pathway

Abstract
Objective:To investigate the protective effect and mechanism of Bufei Decoction(BFD)on acute bronchitis in rats by modulating the PI3K/Akt/HIF-1α/VEGFA signaling pathway.Methods:A rat model of acute bronchitis was established using smoke exposure.SPF-grade male SD rats were randomly divided into six groups(n=10 per group):control group,model group,low-dose BFD group,medium-dose BFD group,high-dose BFD group,and western medicine group.After the treatment period,HE staining was performed to observe pathological changes in bronchial and lung tissues.ELISA kits were used to measure the levels of TNF-α,IL-6,IL-8,IL-10,SOD,MDA,and NO in lung tissue.Western blotting was employed to detect the protein expression of PI3K,p-PI3K,Akt,p-Akt,HIF-1α,and VEGFA in lung tissue.Results:Compared with the control group,the model group exhibited extensive inflammatory cell infiltration in bronchial and lung tissues,significantly increased levels of inflammatory cytokines TNF-α,IL-6,and IL-8(P<0.05),decreased IL-10 level(P<0.05),reduced SOD and NO content,increased MDA content(P<0.05),and upregulated expression of p-PI3K/PI3K,p-Akt/Akt,HIF-1α,and VEGFA(P<0.05).Compared with the model group,the medium-dose and high-dose BFD groups,as well as the western medicine group,showed significant improvement in pathological changes of bronchial and lung tissues and in all the aforementioned indicators(P<0.05).Conclusion:BFD can reduce the secretion of inflammatory factors and enhance antioxidant capacity in the lung tissue of rats with acute bronchitis,which may be associated with its inhibitory effect on the PI3K/AKT/HIF-1α/VEGFA signaling pathway.关键词
