摘要

Abstract

Objective:To investigate the effect of acupuncture on neurological function in mice with ischemic stroke by regulating the miR-372-3p/thioredoxin-interacting protein(TXNIP)axis.Methods:Ischemic stroke mice were successfully constructed by thread embolism method and were divided into the following groups:model group,acupuncture group,acupuncture+miR-372-3p antagonist negative control group(acupuncture+NC group),and acupuncture+miR-372-3p antagonist group(acupuncture+miR-372-3p group).A sham operation group was included,in which mice underwent carotid artery isolation without insertion or ligation.Y-maze and step-through tests were used to assess learning and memory abilities.TTC staining was performed to measure cere-bral infarct area.HE staining was used to evaluate morphological changes in hippocampal neurons.ELISA was em-ployed to detect the expression of inflammatory factors in brain tissue.qRT-PCR was used to measure the expres-sion levels of miR-372-3p and TXNIP mRNA in brain tissue.Western blotting was conducted to assess TXNIP protein expression in brain tissue.Dual-luciferase reporter assays were performed using mouse hippocampal neu-rons to verify the targeting relationship between miR-372-3p and TXNIP.Results:Compared with the sham oper-ation group,the model group exhibited impaired learning and memory,increased cerebral infarct area,significant pathological damage to hippocampal neurons,elevated expression of TXNIP mRNA,TXNIP protein,TNF-α,IL-1β,and IL-6 in brain tissue,and reduced expression of miR-372-3p(P<0.05).Compared with the model group,the acupuncture group showed improved learning and memory,reduced cerebral infarct area,alleviated pathologi-cal damage to hippocampal neurons,decreased expression of TXNIP mRNA,TXNIP protein,TNF-α,IL-1 β,and IL-6 in brain tissue,and increased expression of miR-372-3p(P<0.05).Compared with the acupuncture group or the acupuncture+NC group,the acupuncture+miR-372-3p group exhibited impaired learning and memory,increased cerebral infarct area,significant pathological damage to hippocampal neurons,elevated expression of TXNIP mRNA,TXNIP protein,TNF-α,IL-1β,and IL-6 in brain tissue,and reduced expression of miR-372-3p(P<0.05).Dual luciferase report showed that miR-372-3p targeted and negatively regulated the expression of TXNIP.Conclusion:Acupuncture may up-regulate the expression of miR-372-3p and down-regulate the expres-sion of TXNIP,thereby weakening the neuroinflammatory reaction and improving the neurological function in isch-emic stroke mice.
