三江牛MSTN基因多态性与体尺性状的关联分析研究
Polymorphisms of MSTN Gene and its Correlation with Body Size Traits in Sanjiang Cattle

Abstract
MSTN gene was identified as one of the candidate genes affecting mammalian growth,which was mainly highly expressed in skeletal muscle and closely related to animal growth performance.The aim of this study was to explore the polymorphisms of MSTN gene to provide genetic markers for selection of a high-quality Sanjiang cattle.In this study,phenol chloroform method was used to extract the genomic DNA of 71 Sanjiang cattle,and the single nucleotide polymorphisms of MSTN gene were identified by PCR technique combined with Sanger sequencing method,and subsequently genotyping and genetic polymorphism analysis were performed.The results showed that there were two SNPs on the third exon of MSTN gene of Sanjiang cattle,which were g.6283935A>C(genotypes AA,AC,CC)and g.6283941C>T(genotypes CC,TC,TT),respectively.A SNP locus g.6284013C>A(genotype CC,AC,AA)was found in the non-coding region.The genotype and gene frequencies were the same for these three loci(For example,locus the genotype frequencies of g.6283935A>C were AA(57.75%),AC(38.03%),CC(4.22%),respectively;its gene frequencies were A(76.76%)and C(23.24%),respectively;its homozygosity(Ho),expected heterozygosity(He),effective allele number(Ne)and polymorphic information content(PIC)were 0.643、0.357、1.55 and 0.293,respectively).These three loci were moderately polymorphic(0.25 ≤PIC<0.50)and consistent with Hardy-Weinberg equilibrium(P>0.05).ACC was the dominant haplotype and CCA was the inferior haplotype by the linkage imbalance analysis.Further analysis of coding characteristics showed that the three polymorphic loci in the coding region of gene were synonymous mutations,but the bias of codon was different.These two SNP loci were associated with the trait of breast width in Sanjiang cattle.The above results revealed three SNP loci of MSTN gene,associated with breast width in Sanjiang cattle and provided molecular markers for the breeding of growth traits in Sanjiang cattle.关键词
