大别山牛瘤胃大小比较及其相关调控基因表达分析
Comparison of Rumen Size and Expression of Related Regulatory Genes in Dabieshan Cattle

Abstract
The purpose of this study was to analyze the key regulatory genes and molecular mechanisms of rumen size in Dabieshan cattle,and to provide a theoretical basis for the improvement of local cattle germplasm.Twenty 24-month-old Dabieshan cows with similar weight were selected,and the pre-slaughter live weight and rumen weight were determined.According to the rumen weight and its proportion,3 in each of the high rumen weight group(H)and the low group(L)were screened,and rumen tissue samples were collected.Transcriptome sequencing was performed using the Illumina Hiseq 2000 platform.The results showed that the rumen weight and the proportion of pre-slaughter live weight in group H were significantly greater than those in group L(P<0.05),and there was no significant difference in pre-slaughter live weight between the two groups(P>0.05).A total of 449 differentially expressed genes were identified in the two groups,of which 348 were up-regulated in group H and 101 were down-regulated.GO functional annotation showed that differentially expressed genes were significantly enriched in items such as metabolism,immunity,catalytic activity,transporter activity,transcription factor binding,etc.KEGG pathway enrichment analysis showed that differentially expressed genes were significantly enriched in immune-related pathways,metabolic function regulation pathways,circadian rhythm regulation pathways,and pathways involved in cell tissue structure and function.Based on PPI network analysis and degree algorithm,five key node genes,PTGS2、BCL2、GRO1、IL6R and INSR,were identified as possibly involved in the regulation of rumen immunity-metabolism-tissue structure in Dabieshan cattle.The study compared and analyzed the rumen transcriptome of Dabieshan cattle with different rumen sizes,established a regulatory network related to the rumen immunity-metabolism-tissue structure of Dabieshan cattle,and screened out five core genes of PTGS2、BCL2、GRO1、IL6R and INSR that may co-regulate rumen development.关键词
