Abstract

Objective:To explore the sex identification method of the original animal Eupolyphaga sinensis of Eupolyphaga Steleophaga due to the issue of mixed use in medicinal products.This study aims to provide a reference for improving the quality control of these medicinal materials.Methods:Morphological observation,including macro-traits,micro-traits,and microscopic examination,was performed on male and female larvae and adults of E.sinensis.DNA barcodes-cytochrome oxidase Ⅰ(COI),cytochrome b gene(Cytb),18S rRNA,and 16S rRNA-were used for species identification and sequence comparison between male and female samples across multiple batches.Results:The complete morphological characteristics of male and female E.sinensis larvae and adults were delineated.When E.sinensis specimens were intact,male and female larvae could be distinguished by the number of abdominal transverse annuli and the size of the genital operculum,while adults exhibited clear dimorphism of external features such as the presence or absence of wings.In the identification by DNA barcoding,all samples were accurately identified as E.sinensis,while there was no differential sites in the barcode between male and female individuals.Conclusion:The sex of E.sinensis can be identified by conventional identification techniques when the individual morphology is intact.However,conventional DNA barcoding has limitations for sex identification.Therefore,it is necessary to further study and develop a molecular method for sex identification of E.sinensis.
