基于PI3K/AKT信号通路探讨木香烃内酯对卵巢癌细胞的影响
Exploration of the effect of costunolide on ovarian cancer cells based on the phosphatidylinositol 3-kinase/protein kinase B signaling pathway

Abstract
Objective To investigate the effects of costunolide on the proliferation,migration,and invasion of ovarian cancer A2780 and SKOV3 cells based on the phosphatidylinositol 3-kinase/protein kinase B(PI3K/AKT)signaling pathway.Methods Ovarian cancer A2780 and SKOV3 cells were used as the research objects,and the blank control group,the low,medium,and high concentration groups of costunolide,and the positive control group(PI3K inhibitor group)were set up respectively.CCK-8 assay was used to detect the effect of costunolide on the proliferation of ovarian cancer A2780 and SKOV3 cells;Scratch assay was used to detect the effect of costunolide on the migration of ovarian cancer A2780 and SKOV3 cells;Transwell assay was used to examine the effect of costunolide on the invasion of ovarian cancer A2780 and SKOV3 cells.The expression levels of PI3K/AKT signaling pathway-related proteins p-PI3K,PI3K,p-AKT and AKT were detected by Western Blot.Results Compared with the blank control group,the costunolide treatment group showed inhibitory effects on the proliferation,migration,and invasion of ovarian cancer A2780 and SKOV3 cells in a concentration-dependent manner(P<0.05).Western blotting experiment results showed that the protein expression levels of p-PI3K/PI3K and p-AKT/AKT in the costunolide treatment group were lower than those in the blank control group(P<0.05).There were no statistically significant differences in p-PI3K/PI3K and p-AKT/AKT in the high concentration group of costunolide compared with the positive control group(PI3K inhibitor group)(P>0.05).Conclusion Costunolide may inhibit the proliferation,migration,and invasion of ovarian cancer cells by suppressing the PI3K/AKT signaling pathway.关键词
