摘要

Abstract

Objective To study the mechanism of modified Wugong Baidu decoction in alleviating psoriasis recurrence by regulating autophagy-related pathways.Methods 30 C57BL/6 mice were randomly divided into the blank group,the model group,the western medicine group(0.003mg·mL-1),low-dose group(0.1g·mL-1),medium-dose group(0.2g·mL-1)and high-dose group(0.4g·mL-1).A recurrent psoriasis mouse model was establilshed through twice induction with imiquimod and then administered by gavage.The mice psoriasis area and severity index(MPASI)method was used to evaluate the skin lesion index of each group,and calculate the change in △ MPASI before and after gavage.The histopathological changes of skin lesions in each group were observed by HE staining.RT-PCR method was used to detect the transcription of genes such as adenosine monophosphate activated protein kinase(AMPK),tuberous sclerosis complex(TSC1/2),Ras homolog enriched in brain(Rheb)and mammalian target of rapamycin(mTOR)in skin lesion tissues.The correlation between the key AMPK/TSC/mTOR pathway mediating autophagy and△ MPASI of macroscopic skin lesion was analyzed.Results During the modeling process,it was found that the appearance time of psoriasis-like skin lesions in mice was earlier during the second induction of imiquimod,and the skin lesions were more typical and the manifestations were more severe.The MPASI scores of mice in the drug groups were significantly reduced(P<0.01),with obvious alleviation of epidermal keratinization,recovery of the spinous and granular layers,and reduction of inflammatory cell infiltration under microscopic pathological conditions.In regulating the AMPK/TSC/mTOR pathway,recurrent psoriasis mice exhibited high expression of AMPKα and TSC1,and low expression of TSC2,Rheb and mTOR;And the drugs significantly downregulated AMPKα and TSC1,and upregulated the transcription levels of TSC2,Rheb and mTOR genes in skin lesions(P<0.01).Moreover,the gene expression levels of various factors were correlated with △ MPASI score(P<0.05).Conclusion Modified Wugong Baidu decoction inhibits autophagy by regulating the AMPK/TSC/mTOR pathway,which may indirectly block the energy source of tissue-resident memory T cells(TRM)and prevent the recurrence of psoriasis.
