Genetic diversity analysis of Rhizobia isolated from alfalfa in different ecological area of Anhui Province

摘要

Abstract

[Objectives]With the development of the herbivorous animal industry in southern China,the culti-vated area of alfalfa has increased significantly.Screening Rhizobia meliloti with high nitrogen-fixation efficiency and strong nodulation competitiveness is essential for promoting the sustainable development of the alfalfa industry.[Method]The study isolated strains of Rhizobia meliloti from different ecological regions in Anhui.The 16S rRNA and housekeeping genes(atpD and gln Ⅱ)were sequenced to analyze the genetic evolution,classification,and genetic diversity of natural populations from these regions.[Result]A total of 21 Rhizobia(including 19 strains of Sinorhizo-biumn meliloti)strains were isolated from alfalfa nodules in different regions of Anhui.Sequence analysis revealed 67,49,and 125 polymorphic sites in the 16S rRNA,atpD,and gln Ⅱ nucleic acid sequences,respectively.Among these,gene of gln Ⅱ showed the highest polymorphism index and the greatest average nucleotide divergence.Neutral testing using Fu and Li's D indicated that 16S rRNA conformed to neutral drift,whereas both housekeeping genes were in-fluenced by environmental selection pressures.Genetic distance analysis shows inconsistency among the three gene analysis strains,with 16S rRNA exhibiting the smallest genetic distance,and gln Ⅱ the largest.Phylogenetic cluster-ing based on the three genes yielded broadly consistent results,though minor differences were observed.Notably,Rhi-zobia from the southern Yangtze River region and mountainous areas of Anhui were clustered and separated into dif-ferent genetic subgroups.[Conclusions]All three gene sequences of the isolated strains differed from reference se-quences available in NCBI,suggesting that these isolates may indicate new strains of Sinorhizobium meliloti.
