Comparison of chloroplast genome characteristics and phylogenetic analysis of five Medicago species in Medicago Sect.M

Abstract
[Objective]The aim of this study is to analyze the chloroplast genomic characteristics of five Medi-cago plants in the M.Sect.Medicago group,providing a theoretical basis for the study of the phylogenetic relationship of Medicago group plants.[Method]The chloroplast genomes of four cultivars and M.sativa subsp.caerulea were sequenced by Illumina NovaSeq 6000 platform,and these five newly sequenced plants with 12 Medicago derived from NCBI were phylogenetically analyzed.[Result]The total length of the genome was 121 383 to 127 679 bp,en-coding 108 to 110 genes.All of them lacked the rps16,rpl22 and infA genes.Among them,alfalfa(M.sativa subsp.caerulea)also lacked the clpP and trnI-CAU genes.The total GC content was:33.8%to 33.9%.The repeated se-quences were determined and eight highly variable regions were screened out.Phylogenetic analysis showed that M.falcata and M.falcate'Hulunbeier'were sisters to each other,M.sativa'Deqin',M.sativa'qingshui',M.sativa'Baoding',Medicago sativa subsp.caerulea and M.sativa clustered into a single branch,indicating that they were closely related and had similar genetic background.[Conclusion]The chloroplast genomes of five newly sequenced plants belong to the IRLC group,and the genome structure is similar with a certain diversity.The phylogenetic rela-tionship of these 17 plants in M.Sect.Medicago was supported by the whole chloroplast genome sequence analysis to some extent.关键词
