摘要

Abstract

Soybean Cercospora leaf spot(Cercospora sojina Hara)is a globally prevalent disease and a major constraint in soybean production in China,with severe outbreaks significantly compromising both yield and quality.The soybean cultivar Heinong 531 exhibits high resistance to Cercospora leaf spot under field conditions.To elucidate the resistance characteristics of Heinong 531 against this disease,23 soybean germplasm resources with varying resistance levels-including highly resistant,moderately resistant,susceptible,and highly susceptible materials as controls-were selected for systematic analysis through artificial inoculation with Cercospora sojina.Simultaneously,SSR molecular markers were employed to genotype known disease resistance gene loci,investigating the correlation between genotype and disease resistance phenotype.At the physiological and biochemical levels,key physiological indicators were measured to analyze their relationship with resistance.Additionally,quantitative real-time PCR(qRT-PCR)was used to examine the expression patterns of disease resistance-related genes.The results showed that Heinong 531 had a disease index of 2.2,indicating a highly resistant phenotype.Molecular marker analysis revealed that Heinong 531 carried the resistance-specific allele at the Satt565 locus.Compared with highly susceptible materials,Heinong 531 exhibited significantly increased activities of superoxide dismutase,catalase,and polyphenol oxidase.Furthermore,after inoculation with the pathogen,the expression levels of resistance-related genes such as GmRLK,GmTGA06,and GmSOD1 were significantly up-regulated in Heinong 531.This study provides critical evidence supporting the value of Heinong 531 in disease resistance breeding and contributes to both theoretical and germplasm resources for the development of soybean varieties resistant to frogeye leaf spot.
