The transcriptional regulation of the caffeine degradation gene cluster by CdnR2 in Paraburkholderia caffeinilytica CF1

Abstract
A caffeine degrading gene cluster was discovered in the genome of a highly efficient caffeine degrading bacterium Paraburkholderia caffeinelytica CF1 isolated early in the laboratory.A transcription regulatory factor cdnR2 belonging to the LysR family was found on the gene cluster.The regulatory function of transcription regulatory factor cdnR2 was analyzed in the caffeine degradation of strain CF1.The regulatory function of cdnR2 caffeine degradation in strain CF1 was analyzed by constructing mutants,qRT-PCR and EMSA.The results indicated that CdnR2 inhibited the transcription of cdnR1 by directly binding to PR1 and PK,thereby promoting the transcription of cdnI,cdnJ and cdnK,and indirectly regulating the transcription of cdnH.In addition,caffeine and its degradation intermediates enhance the binding affinity of CdnR2 to PR1 and PK.关键词
