摘要

Abstract

Discoidin domain receptor 1(DDR1)is a distinctive member of the receptor tyrosine kinases(RTKs)family that interacts specifically with collagens,the major components of the extracellular matrix(ECM),to exert diverse biological functions.DDR1 not only participates in physiological processes such as maintaining tissue homeostasis and regulating cellular differentiation but also plays a crucial role in pathological conditions,including fibrosis,inflammation,and tumorigenesis.It is highly expressed in multiple malignancies,such as hepatocellular carcinoma,pancreatic cancer,glioma,breast cancer,and melanoma,and its overexpression correlates with poor prognosis.DDR1 contributes to tumor progression through several mechanisms:it promotes tumor cell survival,proliferation,and differentiation;remodels the ECM;regulates metabolic reprogramming;and maintains cancer stemness.In addition,DDR1 can mediate immune evasion and exert immunosuppressive effects across various tumor types.Currently,several DDR1-targeted drugs have entered clinical and preclinical research.A comprehensive understanding of DDR1's molecular mechanisms and its involvement in tumor biology,coupled with a global landscape analysis of DDR1-targeted drug development,will provide a foundation for developing precision diagnostic tools and targeted therapeutic strategies.The growing body of research on DDR1 highlights its potential as a novel and promising target for cancer therapy.
