复方茵陈蒿汤通过TRPV3/TLR2/NF-κB信号通路抑制痤疮丙酸杆菌诱导的皮肤炎症的机制研究
Fufang Yinchenhao Decoction inhibits Cutibacterium acnes-induced skin inflammation via TRPV3/TLR2/NF-κB signaling pathway

Abstract
Objective:To investigate the effect of Fufang Yinchenhao Decoction on Cutibacterium acnes(C.acnes)-induced in-flammatory response in macrophages by regulating the TRPV3/TLR2/NF-κB signaling pathway,and to elucidate its mechanism in treating acne.Methods:An in vitro cell model of C.acnes infection was established by stimulating macrophages with peptidogly-can,which was then treated with Fufang Yinchenhao Decoction or isotretinoin.The levels of nitric oxide(NO),inflammatory fac-tors(TNF-α,IL-1β,iNOS,and COX2),and oxidative stress markers(SOD,MDA,and ROS)were measured.The expres-sion of key molecules in the TRPV3/TLR2/NF-κB pathway was detected using Western blot and RT-qPCR.Results:Compared to the model group,Fufang Yinchenhao Decoction significantly inhibited the production of NO in macrophages,reduced the levels of TNF-α,IL-1β,iNOS,COX2,and MDA,suppressed the fluorescence intensity of ROS,and enhanced the activity of SOD,with all differences being statistically significant(P<0.01).Western blot and RT-qPCR results further demonstrated that Fufang Yinchenhao Decoction significantly downregulated the expression of TRPV3 protein and mRNA,inhibited the expression of TLR2 and p-IκBα proteins,decreased the ratios of p-IKKβ/IKKβ and p-p65/p65,and upregulated the expression of IκBα protein,with all differences showing statistical significance(P<0.01).Subcellular localization analysis revealed that Fufang Yinchenhao Decoction increased the expression of cytoplasmic p-p65 while reducing the level of nuclear p-p65,with both changes being statisti-cally significant(P<0.01).Conclusion:By inhibiting the activation of the TRPV3/TLR2/NF-κB signaling pathway and regulat-ing oxidative stress and inflammatory responses,Fufang Yinchenhao Decoction exerts its anti-acne effects.关键词
