脊蛇祛湿胶囊对类风湿关节炎合并骨质疏松症患者骨保护作用机制的网络药理学及回顾性研究
The bone-protective mechanisms of Jishe Qushi Capsules in patients with rheumatoid arthritis complicated by osteoporosis:A network pharmacology and retrospective study

Abstract
Objective:To investigate the bone-protective mechanisms of Jishe Qushi Capsule in patients with rheumatoid ar-thritis(RA)combined with osteoporosis using network pharmacology and molecular docking techniques,and to verify these find-ings through a retrospective analysis.Methods:The active ingredients and corresponding targets of the drug were obtained through the TCMSP,BATMAN-TCM,SwissTargetPrediction databases,and relevant literature.Disease-related targets were identified from the GeneCard,OMIM,and TTD databases.Intersection targets were determined using Venny 2.1.0,and the protein-protein interaction(PPI)network was constructed using Cytoscape 3.7.2 and the STRING database.Gene Ontology(GO)functional and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses were performed using the DAVID database.Molecular docking of the main active components and targets was conducted using AutoDock Vina 1.1.2.A retrospective analysis was carried out on patients with RA combined with osteoporosis who were treated at the Second Affiliated Hospital of Guizhou University of Traditional Chinese Medicine from January 2018 to April 2024.The control group(n=81)and the experimental group(n=81)were compared in terms of general information and bone metabolism markers before and after treatment.Results:Jishe Qushi Capsule contained 142 active ingredients,which acted on key targets such as TP53,STAT3,AKT1,SRC,and JUN,exerting bone-protective effects by regulating the PI3K-Akt and MAPK signaling pathways.Molecular docking results showed that the main active components had strong binding affinities with core targets.The retrospective analysis demonstrated that the Δ25-(OH)D3 levels were significantly higher in the experimental group compared to the control group(P<0.05).Furthermore,the combination of"Jishe Qushi Capsule,Alfacalcidol Soft Capsule,Calcium Carbonate and Vitamin D3 Tab-let,and Vitamin D2 Injection"was found to be the most effective regimen(P<0.05).Conclusion:Jishe Qushi Capsule may have a potential bone-protective effect in RA patients with osteoporosis and may produce synergistic effects when combined with anti-osteoporotic medications.关键词
