二黄散对大鼠实验性牙周炎的治疗作用研究
Study on the therapeutic effect of Erhuang powder on experimental periodontitis in rats

Abstract
Objective:To investigate the effects of the traditional Chinese medicine compound formulation Erhuang powder and its main components,catalpol and berberine hydrochloride,in the treatment of perio dontitis,aiming to provide tri-al data support for the clinical application of Erhuang powder in the treatment of periodontitis.Methods:Thirty-six 7~8-week-old male Sprague-Dawley(SD)rats were selected and randomly divided into six groups(n=6):the normal group(Group A),the periodontitis group(Group B),the normal saline group(Group C),the catalpol group(Group D),the berberine hydrochloride group(Group E),and the Erhuang powder group(Group F).No treatment was administered to Group A.Exper-imental periodontitis models were established in Groups B~F by using silk ligatures,followed by corresponding drug treat-ments for two weeks.Western blotting(WB)was employed to detect the protein expression levels of C-C motif chemokine lig-and 5(CCL5)and nuclear factor-kappa B(NF-κB)in the gingival tissues of rats in each group.Real-time quantitative poly-merase chain reaction(RT-qPCR)was used to measure the mRNA expression levels of CCL5,NF-κB,TNF-α,IL-1β,and IL-6 in the gingival tissues of rats in each group.Enzyme-linked immunosorbent assay(ELISA)was performed to assess the changes in the levels of inflammatory cytokines TNF-α,IL-1β,and IL-6 in the gingival tissues of rats in each group.Micro-CT scanning was conducted to evaluate the treatment outcomes of periodontitis in the maxillae of rats in each group.Hematoxylin-eosin(HE)staining was used to explore the infiltration of inflammatory cells in the gingival tissues.Results:In the periodonti-tis group,the periodontal ligament fibers were disordered,and the alveolar bone exhibited a moth-eaten appearance.In the treatment groups,the number of inflammatory cells in the periodontal tissues decreased,and the gingival morphology recovered well.The cementoenamel junction-alveolar bone crest(CEJ-ABC)distances in the treatment groups were significantly shorter compared to the periodontitis group(P<0.01).The protein mRNA and protein expression levels of CCL5 and NF-κB in the periodontitis group were significantly higher than those in the normal group,while both the Erhuang powder group and the berberine hydrochloride group were significantly put down(P<0.05).The mRNA expression levels of inflammatory cytokines(TNF-α,IL-1β,IL-6)in the treatment groups were significantly inhibited compared to the periodontitis group,and the Er-huang powder group exerted an appealing potency and significantly reduced,which was consistent with the ELISA results.Conclusion:Erhuang powder and its active components down-regulated CCL5 and NF-κB expression,reduced the release of inflammatory cytokines,inhibited bone resorption and treated periodontitis.关键词
